芝麻繁育特性研究Ⅴ:花粉活力与柱头可授性

中文摘要:比较分析直接观察法、醋酸洋红染色法、I2-KI染色法、无机酸法、MTT染色法、TTC染色法和FDA荧光检测法在芝麻花粉活力上的检测效果,监测芝麻花粉活力的日变化情况,并采用联苯胺-过氧化氢法和不同时间直接授粉法对芝麻柱头可授性的日变化进行研究。结果表明:(1)直接观察法、TTC染色法和FDA荧光检测法不适合芝麻花粉活力的检测,而醋酸洋红染色法、I2-KI 染色法、无机酸法和MTT染色法等4种方法可用于芝麻花粉活力检测,其中,醋酸洋红染色法操作简单、易于观察。(2)芝麻花粉活力的日变化表现为不断下降趋势,在7:30-9:30最高,11:30-17:30不断下降,达显著水平。(3)联苯胺-过氧化氢法测定的芝麻柱头可授性的日变化表现为先升后降,早晨7:30时已有很高的可授性,随后稍有升高, 9:30时达到最高值,11:30后随时间推延呈显著下降趋势。(4)不同时间直接授粉法测得芝麻授粉时间越晚,结蒴率越低,7:30时具有最高的结蒴率;相对结实率在7:30-11:30时差异不明显,均处在较高状态,13:30后开始不断显著下降。(5)花粉活力、柱头可授性、结蒴率和相对结实率等4个指标的日变化值的相关分析结果表明,4个指标相互之间的相关性均呈极显著正相关,其中柱头可授性与相对结实率的相关系数最大,达 0.940。

Study on Breeding Characteristics of Sesamum indicum L. Ⅴ:Pollen Viability and Stigma Receptivity
Abstract:The direct observation method, acetocarmine staining method, I2-KI staining method, inorganic acids treatment method, MTT staining method, TTC staining method and FDA fluorescence detection method wereused to compare and analyzedetection effect in sesame pollen vitality. Thediurnal variations of sesame pollen viability was monitored and the diurnal variation of sesame stigma accessibility was studied by benzidine-hydrogen peroxide method and direct pollination method at different time.The results showed that:(1) direct observation method, TTC staining method and FDA fluorescence detection method were not suitable for detecting sesame pollen viability. The four methods including acetocarmine staining method, I2-KI staining method, inorganic acids treatment method and MTT staining method could be used for detecting sesame pollen viability, among which acetocarmine staining method was asimple and easy way to be observed. (2) the diurnal variation of sesame pollen viability showed a decreasing trend, which was highest from 7:30 to 9:30, and continuously decreased from 11:30 to 17:30, which reached the significant level. (3)the diurnal variation of sesame stigma receptivity measured by benzidine-hydrogen peroxide method showed a trend of risingat first and then falling. The receptivity was high at 7:30 in the morning, then slightly increased, reached the highest value at 9:30, and then it showed a significant decline trend with time delay after 11:30. (4) The results of direct pollination method at different time showed that the later pollination time was, the lower the capsule setting rate was, and the highest capsule setting rate was detected when it was pollinated at 7:30. The relative seed setting rate had high values from 7:30 to 11:30 with no significant difference, and decreased significantly after 13:30. (5) the correlation analysis of four indexes, including pollen viability, stigma receptivity, capsule setting rate and relative seed setting rate, showed significantly positive correlation with each other, and the correlation coefficient between stigma receptivity and relative seed setting rate was the greatest,which reached to 0.940.
